Fluorescent compounds can be used to measure P450 activity. However, many of the currently available test compounds have shortcomings, such as being dependend on further reactions for detection (such as commercially available luciferine derivatives) or being more or less unspecific with respect to different P450 isoenzymes. Also, there are hardly any substrates for non-hepatic P450s. Therefore, the identification of new fluorescent compounds that are metabolized by human P450s is the aim of this subsproject. For the fluorescent compounds project, they can be used as test substrates to measure P450 activity. In the best case, several compounds with different fluorescence properties can be found that can be analyzed together; this would allow us to set up a new multiplexed P450 assay that determines the activity of several human P450s simultaneously. |
