1) Objective:
Screen out the active peptides which can effectively block the interaction of APRIL and its receptors. Construct the peptides-Fc confusion protein to the get more stable and active peptibody.
2) Method:
a) Molecular modeling the BCMA1, TACI, and 814 peptides which is the potential antagonist of APRIL-receptors;
b) Culture and purify the APRIL, BCMA1-Fc, TACI-Fc, 814-Fc protein;
c) Examine the activity of those purified proteins by ELISA;
d) Screen out the active peptides through ELISA;
e) Construction of fusion peptibody
Extract the plasmid enzymes digest the plasmid reconstruct the peptide-plasmid by T4 DNA ligase examine the accuracy of the recombined plasmid transfer the fused plasmid into Ecoli
purify the protein examine the activity of the protein through ELISA.
3) Result:
a) The BCMA1, TACI, 814 peptides can block the interaction of APRIL and receptors.
b) The BCMA1-Fc, TACI-Fc, 814-Fc protein can block the interaction of APRIL and receptors. |
