1) Objective:
Explore the underpinning molecular mechanism of the longevity effect of Emodin.
2) Method:
a) Signaling pathway: determine the functioning signaling pathway of emodin through evaluating its effect on lifespan of mutant worms;
b) Localization of transcription factors: observe the influence of emodin on the nuclear translocation of DAF-16 and SKN-1;
c) Genes transcription: examine the effect of emodin on the transcriptional levels of aging-related genes through RT-PCR assay;
d) Pharyngeal pumping and reproduction: census the pharyngeal pumping times and reproduction quantity of each worm to determine whether emodin functions through Calorie Restriction (CR) or reproduction inhibition.
3) Result:
a) Emodin extense lifespan via insulin-signaling pathway depending on DAF-16 and SIR-2.1;
b) Emodin extends lifespan of C.elegans without influencing pharyngeal pumping and reproduction. |
