Mitochondrial ROS detection of γ-CD/Cor complexes and mPEG-Cor micelles was observed via MitoSOX™ Red mitochondrial superoxide probe by a Zeiss LSM710 confocal laser scanning microscope. After digestion and counting, PC-3 cells were seeded on 20-mm plates at a density of 105 cells per well. After cell adherence, the previous culture medium was discarded, and 1 mL new culture medium containing 100 μM Cor of two formulations was added into every plate for 3 h. After 3 h, the cells were washed by PBS in triplicate and supplemented with 1mL Hankʼs balanced salt solution (HBSS) with calcium and magnesium containing 5 μM MitoSOX™ protected from light. 30 min later, the HBSS was discarded and the cells were gently washed by 1 mL HBSS three times. 1 mL fresh DMEM medium without FBS was added to maintain cell activity. The cells were then irradiated with light irradiation (365 nm, 95 mW cm-2) for 20 min, and their images were observed using a Zeiss LSM710 confocal laser scanning microscope. The excitation and emission wavelength of DHE was 514 nm and 500-600 nm, respectively. |
