题目: 1.Immunohistochemistry method for detecting HIF-1α
内容提要: Elevated HIF expression has been associated with poor prognosis in many cancers and correlates with reduced survival for patients. HIF-1αwas detected by immunohistochemistry. The paraffin-embedded tumors were deparaffinized in xylene and hydrated in a series of graded alcohols. The tumor sections were repaired with sodium citrate buffer (pH=6.0) and blocked with goat serum. The tumor sections were incubated with HIF-1α monoclonal antibody for mouse at 4°C overnight. Afterwards the tumor sections were incubated with Horseradish peroxidase (HRP) labeled goat anti-mouse IgG (H+L) at room temperature for 30 min and were visualized using DAB as chromogen for 10 minutes. The nucleus was stained with hematoxylin follow by gradient dehydration. HIF-1αin tumor sections was observed by microscope.
