α-Glucosidase enzyme inhibitors are clinically used for the treatment of type 2 diabetes mellitus. In rencent years, it is found that the whole plant of Potentilla discolor have been used to treat type 2 diabetes, it can protect pancreatic β-cell on type 2 diabetes, decerase the blood glucose, and improve the insulin resistance in clinical research. We tested α-glucosidase inhibitory effects of extract and some pure compounds from Potentilla discolor. The α-glucosidase inhibitory capacity of different fractions and some compounds from the ethanol extract of the air-dried whole herb of Potentilla discolor were assessed by PNPG (4-nitrophenyl-α-glucopyranoside) method. All of the extracts showed significant inhibition against α-glucosidase enzyme compared to acarbose. PDE extract was found to be the most potent inhitor with an IC50 of 1 μg/mL. Among the tested compounds, the PDC-5, PDC-6, PDC-7, PDC-8 and PDE-8 had stronger α-glucosidase inhibitory effect compared to acarbose, and the PDC-7 showed the greatest inhibition with IC50 3 μg/mL againstα-glucosidase. |
