we screen 8 residues continuously located on FG loop to build the novel fusion protein with FN3 being used as a scaffold. The fibronectin type III domain (FN3) is a small autonomous folding unit which occurs in many animal proteins involving in ligand binding. Many literature has already reported the the potential of FN3 as a scaffold for engineering novel binding proteins. There are 15 repeating units of FN3 in human fibronectin and we choose the tenth FN3 unit of human fibronectin to build our systems. FN3 is small (94 residues) and monomeric whose structure is best described as similar to that of the antibody VH domain suggesting that the FN3 scaffold is an effective framework onto which loops for specific building functions can be grafted.
