Terpenoids are among the most abundant natural products. Many are highly valuable in pharmaceutics and other industries.Terpene synthases found in all kingdoms of lives are responsible for the formation of diverse terpenes in nature. Most terpene synthases are promiscuous in terms of product formation with some extremes capable of producing hundreds of structurally and stereochemically different products simultaneously. Hence the commonly used chromatography-based assays are problematic with messy elution profiles and result in losing tracks or mis-assigning some products. Reliable activity assays require authentic standards with some commercially not available therefore are difficult, and cost-ineffective. Here we used yeast inorganic pyrophosphatase coupled colorimetric assay to quantitatively characterize three santalene synthases and correlate to the GC-MS method. Among the three, SaSSy and SspiSSy are both originated from sandalwood involved in Sandalwood oil biosynthesis and a phylogenetically distant SanSyn is from Clausena lansium (Wampee). |
