First, the standard curve of the drug in a certain concentration range was determined by high performance liquid chromatography. Then use a volumetric flask to prepare certain concentration of organic solution of drug-loaded micelles, and determine the drug content in the liquid phase. The drug loading of drug-loaded micelles can be calculated as the percentage of the drug in the drug-loaded micelles.
