Buckybowls such as corannulene are an interesting class of compounds with many different potential applications. However, nothing is known about their effects on humans or animals. Previous studies in our group failed to unambiguously identify P450 metabolites of corannulene, which in part was due to analytical problems. It is therefore my task in this subproject to study the metabolism of corannulene itself and of eight corannulene derivatives. For corannulene project, we want to know in principle whether buckybowls are metabolized in humans or not. This project is also interesting from a mechanistic point of view, because it is known that the reaction mechanisms for the P450-dependent hydroxylation of either aliphatic or aromatic compounds are different from each other. Since buckybowls have properties that place them between these two classes it will be interesting to see whether (in case any reaction occurs at all) they with respect to hydroxylation by P450s they are closer to one or the other group. |
