We choose naphthalimide derivative as a fluorophore.Because we have known that naphthalimides can pass into cells and allow cellular assays. As for phosphine probes we have two plans.one is use analogs of triphenyl phosphine, which is photostable,another is use tris-carboxyethylphosphine (TCEP),which is used in biochemistry as a cell-permeable reducing agent.We have two ideas for phosphine probes. One idea is to use analogs of triphenyl phosphine, which does not photo-oxidize. 2-Carboxyphenyl- and 4-carboxyphenyl diphenylphosphine are known compounds. We have not had good luck with the literature preparation, and will instead use Knochel’s method for transmetallation. This method allows the formation of stable aryl Grignards in the presence of esters. After hydrolysis, the free carboxylate can be used to attach the phosphines to amino fluorophores using standard coupling coniditions. Two fluorophores will be tested. One is a nitrobenzoxadiazole. It is similar to a literature compound that can get into cells. The other fluorophore is a naphthalimide derivative. We have shown that naphthalimides can pass into cells and allow cellular assays. While these fluorophores are not as bright as fluorescein derivatives, they are much easier to make. They are also more photostable. Both amino fluorophores have been prepared already. |
