Cyanobacteria have the CO2-concentrating mechanism. Under the low concentration of CO2, dissolved inorganic carbon could enter the cytosol through HCO3− transporters. HCO3- in cytosol subsequently is converted to CO2 by enzyme CA, the concentration of CO2 around Rubiso for photosynthesis increases. In order to improve the efficiency of CO2 fixation, we overexpressed the HCO3- transporter gene BicA. The increased carbon flowed to the MEP pathway for the production of limonene under the action of limonene synthase. The increase of CO2 concentration increased the concentration of substrate G3P for limonene. Limonene productivity reached to an average productivity of 885 μg/L/OD/d. |
