In this experiment, we observed a dose-dependent increase of activity, which is likely the result of two opposing effects: The increase of cell permeabilization with rising Triton X-100 concentrations, which facilitates UGT activity by permitting entry of the cofactor UDPGA (as it cannot be produced by strain CAD200 itself and thus needs to be added to the reaction buffer) as well as that of the substrate, products can also get out from the cells easily for the same reason, and the adverse effect on UGT activity described above. In summary, with increasing concentrations the permeabilization effect outweighs the adverse influence of the surfactant, with the highest UGT activity being observed at 0.3% (v/v) Triton X-100. Therefore, this concentration was used from here on throughout. |
